Tolo Harbour is adjacent to The Chinese University of Hong Kong. The view of Tolo Harbour from the mountaintop of the university has a unique charm, especially when seen from the Pavilion of Harmony between Man and Nature at New Asia College, where the beautiful scenery of the harbour unfolds before your eyes, with distant green hills faintly visible and water blending seamlessly with the sky. It is one of the major inland harbours in the New Territories of Hong Kong, located to the north of Sha Tin District and to the east of Tai Po District. The harbour runs in a southwest-northeast direction, with its outlet at the Chek Mun Channel, where seawater flows into Tolo Harbour and then into Mirs Bay in northeastern Hong Kong. Several rivers flow into Tolo Harbour, including the Shing Mun River in Sha Tin and the Lam Tsuen River in Tai Po.

In the area of Tai Mei Tuk to the north of Tolo Harbour lies Plover Cove, which was once a large bay within the harbour. In the 1960s, due to water shortages in Hong Kong, the government built a dam to completely enclose the bay, pumped out the seawater, and turned it into Plover Cove Reservoir. There used to be several villages along the shores of Plover Cove, which were relocated during the construction of the reservoir.

In the 1980s, to improve transportation between northern New Territories and Sha Tin, the government constructed Tolo Highway along the southern shore of Tolo Harbour between Sha Tin and Tai Po. In 2000, the government expanded Tolo Highway and reclaimed land in the Ma Liu Shui area to build the Hong Kong Science Park.
